Precautions for Antigen Heat Retrieval in Paraffin-Embedded Tissue Sections
1. Do not use copper staining racks to prevent affecting antigen retrieval effectiveness due to changes in the retrieval solution’s pH value;
2. After retrieval is complete, the sections must not be removed until the retrieval solution has cooled. It is recommended to use a gentle cooling method rather than rapid cooling, as rapid cooling can easily cause tissue detachment and insufficient antigen exposure;
3. After removal, the sections need to be thoroughly washed with tap water; otherwise, it may adversely affect subsequent immunohistochemical staining results;
4. To prevent tissue detachment, slides must be treated with an anti-detachment coating before use (Mai Xin product number: SLI-2001);
5. The amount of retrieval solution must be sufficient to immerse all sections;
6. The retrieval time can be adjusted accordingly based on the specific conditions of each laboratory.
