Method of Using Counterstain Solution

2. Scope of Application: Hematoxylin is suitable for counterstaining after AEC, DAB, AP Red / Fast Red, or immunogold-silver staining. Methyl Green is suitable for counterstaining after DAB or BCIP/NBT staining. Nuclear Fast Red is suitable for counterstaining after BCIP/NBT or immunogold-silver staining.
3. Product Form: Liquid, ready-to-use, packaged as 6ml × 3.
4. Instructions for Use:
(1) When using Hematoxylin, apply two drops onto the tissue section. The staining time varies from 30 seconds to 2 minutes; please adjust according to the actual situation and needs. After counterstaining, rinse with tap water. Differentiation can be performed with 0.1% hydrochloric acid, followed by rinsing with ammonia water (0.1%) or PBS (pH 7.4) to return to blue until the nuclei are stained blue.
(2) When using Nuclear Fast Red, apply two drops onto the tissue section. The staining time varies from 30 seconds to 5 minutes; please adjust according to the actual situation and needs. After counterstaining, rinse with tap water. The nuclei will be stained red, and the cytoplasm will appear light pink.
(3) When using Methyl Green, apply two drops onto the tissue section and stain for 5-10 minutes. Rinse under running water for 1-2 minutes until the nuclei turn green.
5. Precautions:
(1) The kit should be stored at room temperature with a shelf life of 1 year.
(2) In immunohistochemistry experiments, if staining such as AEC, AP Red/Fast Red, or BCIP/NBT is used, after counterstaining, mounting can be done directly with an aqueous mounting medium (e.g., AEC mounting medium) without dehydration and clearing. If staining such as DAB is used, after counterstaining, gradient alcohol dehydration and xylene clearing should be performed before mounting with neutral balsam.
