Immunohistochemical Staining Methods
One of the immunohistochemical chromogenic methods
DAB Chromogen
1、Scope of Application: Suitable for immunohistochemical staining using the horseradish peroxidase (HRP) detection system.
2. Product Number: DAB-0031/1031
3. Product Form: Liquid. Contains the following components:
A: DAB Buffer (20×)
B: DAB Substrate (20×)
C: DAB Chromogen (20×)
4. Usage Method:
⑴ Preparation Method: Taking the preparation of 1 mL of chromogenic solution as an example: Add 50 μL each of reagents A, B, and C to 0.85 mL of distilled water, and mix well (the prepared solution should be stored away from light and is effective within 30 minutes);
⑵ Apply an appropriate amount of the prepared chromogenic solution onto the tissue section. Stain at room temperature for 2-10 minutes. The staining time can be monitored under a microscope. The staining result is brownish-yellow (brown).
Immunohistochemical Chromogenic Method Two
Enhanced DAB Chromogen
1. Scope of Application: Suitable for immunohistochemical staining using the horseradish peroxidase (HRP) detection system.
2. Product Number: DAB-2031/2032
3. Product Form: Liquid. Contains the following components:
A: DAB Buffer (Ready-to-Use)
B: DAB Substrate (20×)
C: DAB Chromogen (20×)
4. Usage Method:
⑴ Preparation Method: Add 50 μL each of reagents B and C to 1 mL of reagent A, and mix well (the prepared solution can be stored away from light at 2°C-8°C for up to one week, with chromogenic effect consistent with freshly prepared DAB);
⑵ Apply an appropriate amount of the prepared chromogenic solution onto the tissue section. Stain at room temperature for 2-10 minutes. The staining time can be monitored under a microscope. The staining result is brownish-yellow (brown).

B: DAB Chromogen (20×)
4. Usage Method:
⑴ Preparation Method: Add 50 μL of reagent B to 1 mL of reagent A, and mix well (the prepared solution can be stored away from light at 2°C-8°C for over two weeks, with chromogenic effect consistent with freshly prepared DAB);
⑵ Apply an appropriate amount of the prepared chromogenic solution onto the tissue section. Stain at room temperature for 2-10 minutes. The staining time can be monitored under a microscope. The staining result is brownish-yellow (brown).

AEC Chromogen
1. Scope of Application: Suitable for immunohistochemical staining using the horseradish peroxidase (HRP) detection system.
2. Product Number: AEC-0037
3. Product Form: Liquid. Contains the following components:
A: AEC Buffer (20×)
B: AEC Substrate (20×)
C: AEC Chromogen (20×)
4. Usage Method:
⑴ Preparation Method: Taking the preparation of 1 mL of chromogenic solution as an example: Add 50 μL each of reagents A, B, and C to 0.85 mL of distilled water, and mix well (the prepared solution should be stored away from light and is effective within 30 minutes);
⑵ Apply an appropriate amount of the prepared chromogenic solution onto the tissue section. Stain at 37°C for 10-30 minutes. The staining time can be monitored under a microscope. The staining result is red.

BCIP/NBT Chromogen
1. Scope of Application: Suitable for immunohistochemical staining using the alkaline phosphatase (AP) detection system.
2. Product Number: NBT-2200/2211/2222
3. Product Form: Liquid, ready-to-use.
4. Usage Method: Apply an appropriate amount of chromogenic solution onto the tissue section. Stain at room temperature for 10-30 minutes. The staining time can be monitored under a microscope. The staining result is blue-black.
